Abstract

Ciprofloxacin (CIP) is a broad-spectrum antibiotic effective against Gramnegative organisms in clinical and hospital settings. However, bacteria strains resistant to it have emerged in recent years. This study assesses the bacterial community in hospital wastewater treatment plants (WWTPs) and investigates the impact of CIP treatment. Next-generation sequencing targeting the 16S rRNA genes was conducted on wastewater samples (A) and samples treated with 10 mg/L CIP using different growth media: Nutrient broth (rich medium) (B) and a minimum salt medium (poor medium) (C). Decrease in
Chao1, Pielou, observed OTUs, Shannon, and Simpson indices indicated reduced species richness and diversity. Microbial communities in treatments A, B, and C showed notable
dissimilarities between each other. Bray-Curtis and weighted UniFrac PCoA plot explained 54.86% and 74.32% of the total variance, respectively. Analysis across samples revealed
that Firmicutes exhibit the highest relative frequency of bacteria (40.62%), followed by Proteobacteria (35.19%) and Bacteroidetes (19.92%). Barnesiellaceae dominate treatment
B and Sphingobacteriaceae prevail in treatment C at the family level. The CIP-resistant bacteria in different treatments shape the microbial community because of their adaptability
to the ecosystem. Therefore, it is crucial to consider these ecological implications for environmental bioremediation and development.
